BT-483 breast cancer cell line shows pleomorphic nuclei and epithelial features, serving as a reliable model for breast carcinoma research studies.
BT-483 is a human epithelial breast carcinoma cell line originating from solid ductal invasive carcinoma tissue (ER⁺/HER2⁻). The cells display pleomorphic, epithelial-like morphology, featuring prominent nuclei and nucleoli, desmosomes, microvilli, tight and gap junctions. Cytogenetic analysis reveals an aneuploid karyotype with a modal chromosome count around 72. BT-483 harbors an activating PIK3CA E542K mutation, which drives elevated PI3K/AKT pathway activity independent of HER2 expression. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

BT-483 serves as a robust in vitro model for hormone-responsive breast cancer and is widely used in BT 483 experiments investigating estrogen receptor signaling and PI3K-driven tumor growth. Under optimized BT 483 culture conditions, these cells are also employed in functional HER2/PI3K pathway assays—particularly the CELx HSF test—to evaluate drug responses to HER2-targeted therapies such as pertuzumab and lapatinib, due to their unusually high HER2 pathway activity despite being classified as HER2-negative.
