The Dual One Step Luciferase Reporter Gene Assay Kit is a dual-luciferase reporter gene assay featuring high sensitivity and a stable luminescent signal.
Firefly luciferase is a ~61 kDa enzyme that catalyzes the oxidation of luciferin to oxyluciferin in the presence of ATP, magnesium ions, and oxygen, producing bioluminescence with a peak emission at approximately 560 nm. Renilla luciferase is a smaller enzyme (~36 kDa) that catalyzes the oxidation of coelenterazine to coelenteramide in the presence of oxygen, emitting light at around 480 nm. The Dual One Step Luciferase Reporter Gene Assay Kit is a glow-type dual-luciferase assay that provides high sensitivity and a stable luminescent signal. The kit contains highly purified luciferin and coelenterazine, allowing sequential detection of firefly luciferase followed by Renilla luciferase within the same sample. The firefly signal is first measured and then quenched, enabling simultaneous detection of Renilla luciferase as an internal control. This normalization corrects for variations in cell number and transfection efficiency, improving assay accuracy.

Compared with flash-based dual-luciferase assays, this kit offers longer signal stability, simplified add–mix–measure workflows without autosamplers or centrifugation, and no noticeable odor. It is compatible with common cell culture media, including RPMI 1640, DMEM, MEM-α, F12, and DMEM/F12, with a signal half-life of approximately 2 hours at 22°C, making it ideal for high-throughput applications. The interaction between the detection reagent and luciferase produces optimal luminescence intensity. Adding Renilla luciferase substrate allows for the detection of luminescence quenching by firefly luciferin. The assay exhibits normal luminescence with minimal variation in luminescence performance across a variety of culture media.
