The Fixation Buffer contain paraformaldehyde, used to stabilize the membranes, fixed and permeabilized cells prior to intracellular cytokine staining.
Fixatives and permeabilization agents are used to treat cell membranes prior to intracellular cytokine staining. Fixatives stabilize the cell membrane, preserving surface antibody-antigen binding. Permeabilization agents create pores in the fluid, intact cell membrane, facilitating antibody entry. When using flow cytometry to detect intracellular antigens and DNA, cells are fixed and permeabilized before adding the appropriate antibodies or detection reagents and incubating for a specified period before analysis.
