Human Aortic Endothelial Cells-Diabetic Type II model endothelial dysfunction in diabetes, aiding research on cardiovascular complications and therapies.
Human Aortic Endothelial Cells-Diabetic Type II are primary cells isolated from the thoracic aorta of confirmed Type 2 diabetic human donors. Following primary culture, these cells are cryopreserved. These endothelial cells display characteristic cobblestone morphology while demonstrating hallmark metabolic dysfunction, including downregulation of PPARγ and activation of SREBP-1c pathways. They serve as a valuable in vitro model for investigating endothelial repair mechanisms in diabetes and the regulatory role of vascular endothelial growth factors in diabetes. Therefore, they have been identified as critical contributors to diabetic vasculopathy, including accelerated atherosclerosis, hypertension, insulin resistance-related macroangiopathy, and microvascular complications like nephropathy and retinopathy. This population expresses surface markers including CD31, VE-cadherin, and RAGE. The diploid karyotype is maintained in >95% of cells. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

Human Aortic Endothelial Cells-Diabetic Type II serve as a pathophysiologically relevant in vitro model to study diabetic macroangiopathy in conditions like accelerated atherosclerosis and microvascular complications. Their metabolic dysregulation enables study of TNF-α/NF-κB-driven VCAM-1 overexpression and subsequent monocyte adhesion under hyperglycemia. Researchers can utilize this model to evaluate SGLT2 inhibitors, test shear stress-optimized drug-eluting stents and develop therapies targeting plaque stabilization and prevention of diabetic end-organ damage. The preserved insulin resistance phenotype of the cells makes them particularly valuable for translational research bridging molecular mechanisms with clinical interventions.
