Human plateable hepatocytes are primary parenchymal cells isolated from whole human liver tissue, retaining native functionality at an early passage. These polygonal epithelial cells exhibit abundant mitochondria and Golgi apparatus, reflecting their roles in metabolism (e.g., ketogenesis), protein synthesis (e.g., albumin), and detoxification (via cytochrome P450 enzymes). They maintain a normal diploid karyotype, ensuring genetic stability, and express key markers like albumin, CYP450s, and innate immune proteins (Complement C3, MASPs, CRP). In non-alcoholic fatty liver disease (NAFLD), hepatocytes display metabolic dysfunction, including impaired lipid processing. Their dual metabolic-immune functions—such as secreting acute-phase proteins (e.g., CRP)—highlight their centrality in liver homeostasis and disease. These plateable hepatocytes are ideal for in vitro studies of liver physiology, drug metabolism, and disease modeling due to their ability to adhere and maintain functional activity.
