The IL21 Reporter Cell Line is a luciferase reporter gene cell line constructed based on the JAK-STAT signaling pathway.
IL-21 is a cytokine with a four-helix bundle structure. The IL-21 receptor (IL-21R) is a heterodimer consisting of unique subunits and the r chain, widely expressed on the surface of lymphocytes and hematopoietic cells. It shares the γ chain with other members of the IL-2 family (IL-2, IL-4, IL-7, IL-9, IL-13, and IL-15). The IL-21R subunit serves as the ligand recognition binding site, while the γ chain is a signaling transduction unit. Signal transduction of IL-21 mainly occurs through the Janus kinase (JAK)/signal transducer and activator of transcription (STAT) pathway. Upon binding to its receptor, IL-21 activates JAK family members (JAK1 and JAK3), leading to the phosphorylation of STAT1, STAT3, STAT4, and STAT5, which regulate gene expression in the nucleus. Additionally, IL-21 can also activate members of the mitogen-activated protein kinase (MAPK) family.The IL21 Reporter Cell Line is a luciferase reporter gene cell line constructed based on the JAK-STAT signaling pathway. When the IL-21 receptor binds, it activates the STAT signaling pathway, thereby inducing the expression of luciferase. The luciferase reading represents the activation effect of the signaling pathway and can be used to evaluate the in vitro effects of IL-21-related drugs.
