MRC-5 is a human lung fibroblast cell line that was originally isolated from the normal lung tissue of a 14-week-old male fetus. MRC-5 exhibits fibroblast-like morphology and adherent culture properties. This cell line is a normal diploid with a karyotype of 46, XY. The modal chromosome number is 46, with 70% of cells showing this count and a polyploidy rate of 3.6%. MRC-5 cells are non-tumorigenic.

MRC-5 cells enable vaccine production against hepatitis A, varicella, and rabies and support viral research by studying replication/infection mechanisms of poliovirus 1, herpes simplex virus, and SARS-CoV-2 for antiviral development. They facilitate cancer research investigating tumor microenvironments and cancer-fibroblast interactions while offering potential stem cell applications due to fibroblast-like morphology and differentiation capacity. These cells further serve in biopharmaceutical/vaccine manufacturing and provide PCR control material via gDNA products to assess inhibition in bioproduction processes.
