The TUNEL (TdT-mediated dUTP Nick End Labeling) Apoptosis Detection Kit can be used to detect apoptosis (programmed cell death) in individual cells.
During cell apoptosis, cells activate several DNA endonucleases, which cleave genomic DNA between nucleosomes. When DNA is extracted from apoptotic cells and examined by electrophoresis, a 180-200 bp DNA ladder is observed.
The TUNEL (TdT-mediated dUTP Nick End Labeling) Apoptosis Detection Kit (Fluor 488) can be used to detect nuclear DNA fragmentation in tissue cells during the late stages of apoptosis. It works by incorporating Fluor 488-12-dUTP into the 3´-hydroxyl (3´-OH) ends exposed by genomic DNA fragmentation under the action of terminal deoxynucleotidyl transferase (TdT), enabling detection using fluorescence microscopy or flow cytometry.
