C57BL/6 Mouse Primary Mammary Fibroblasts from AcceGen are isolated from tissue of pathogen-free laboratory mice.
C57BL/6 Mouse Mammary Fibroblasts are primary cells isolated from the mammary tissue of pathogen-free laboratory mice. They primarily reside in the stromal compartment of mammary tissue, specifically within the fat pad connective tissue surrounding ducts and lobules. Following primary culture, these cells are cryopreserved. The cells play critical roles in extracellular matrix remodeling (collagen and fibronectin deposition), modulate epithelial-stromal crosstalk via growth factor signaling TGF-β and HGF, and regulate angiogenesis (VEGF secretion). Studies demonstrate their implication in breast cancer, mammary fibrosis, and lactational insufficiency. These mouse fibroblasts are characterized by their spindle-shaped morphology and expression of fibroblast markers FSP1/S100A4. For optimal performance, repeated freezing and thawing should be avoided during culture. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

C57BL/6 Mouse Mammary Fibroblasts serve as an in vitro model to study the pathogenesis of breast cancer stroma in disease conditions such as triple-negative breast cancer (TNBC). Researchers can utilize this system to investigate TGF-β/Smad-mediated cancer-associated fibroblast (CAF) activation in TNBC progression, examine CXCL12/CXCR4 signaling in therapy resistance, and evaluate stromal contributions to extracellular matrix stiffening and immunosuppressive niche formation. These cells are particularly valuable for studying mechanisms of extracellular matrix stiffening, immunosuppressive niche formation, and metastatic vascular co-option.
