COLO-205 Cell Line from AcceGen is a colon adenocarcinoma model widely applied in gastrointestinal cancer biology, drug discovery, and biomarker research.
COLO‑205 is a human colorectal adenocarcinoma cell line derived from the ascitic fluid of a 70-year-old male with Dukes’ stage D colon cancer. The cells exhibit an epithelial-like morphology, growing as both loosely adherent monolayers and suspended spherical aggregates. These markers occur in monosomic, disomic, and trisomic conditions, although most are present as monosomic. COLO‑205 expresses epithelial markers such as keratin and secretes low levels of carcinoembryonic antigen (CEA). These cells are tumorigenic in nude mice, forming moderately differentiated adenocarcinomas that reflect human colon cancer histology. Genetically, they are microsatellite stable (MSS) and harbor mutations in key genes including APC, BRAF, CTNNB1, and TP53. A mutation in E-cadherin contributes to enhanced tumorigenicity and altered adhesion. Although COLO‑205 cells do not readily grow in soft agar, they remain a robust model for studying colorectal cancer biology. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, syphilis, mycoplasma, fungi, yeast, and bacteria.
