Explore HCT-8, a tumorigenic human colorectal adenocarcinoma cell line. This adherent, cytokeratin-positive line is a key model for cancer research.
HCT-8 is a human colorectal adenocarcinoma cell line derived from the ileocecal region of a 67-year-old male patient with intestinal carcinoma. These cells grow adherently in vitro, displaying epithelial-like, polygonal morphology with tight cell-cell junctions, and can be continuously passaged. HCT-8 cells express epithelial markers such as cytokeratins and E-cadherin, consistent with their intestinal epithelial origin. They produce brush border enzymes and transporters typical of intestinal epithelial cells, making them valuable for studying intestinal barrier function, drug absorption, and host-pathogen interactions. At the molecular level, HCT-8 cells harbor mutations commonly found in colorectal cancers, including alterations in KRAS and APC genes. Their responsiveness to inflammatory stimuli and ability to form polarized monolayers contribute to their widespread use in gastrointestinal research and cancer biology.

The HCT-8 cell line is a valuable tool in both cancer and toxicology research. It is widely used to study drug resistance and to elucidate the mechanisms underlying chemoresistance. These cells also support investigations into cell death pathways, including apoptosis and necrosis. Notably, HCT-8 cells are used in the construction of compartmentalized three-dimensional (3D) intestinal models, offering an alternative culture system for continuous cell propagation and enabling detailed studies of gastrointestinal diseases and drug responses.
