DLD-1 is one of two colorectal adenocarcinoma cell lines which were isolated by D.L. Dexter and associates during a period from 1977-1979
DLD-1 is a colorectal adenocarcinoma cell line isolated from the large intestine of a colon adenocarcinoma patient. These cells exhibit an epithelial-like morphology and grow as an adherent monolayer. DLD-1 has a pseudodiploid karyotype with a modal chromosome number of 46, reflecting a structurally altered but numerically near-diploid genome, a pattern often associated with chromosomal instability in cancer cells. They express GCC2, PERP, and IPO7. Each lot undergoes rigorous screening and isolation procedures, and is rigorously tested to ensure it is free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

DLD-1 is extensively employed in colorectal cancer (CRC) research due to its well-characterized genetic and phenotypic profile. As a microsatellite instability-high (MSI-H) model, it serves as a valuable tool for studying DNA mismatch repair defects and their implications in tumorigenesis. DLD-1 is widely used in drug sensitivity and resistance assays, particularly for evaluating chemotherapeutic agents and targeted therapies, such as EGFR or BRAF inhibitors.
