HT-29 cell line supports colorectal cancer research by providing a human adenocarcinoma model for differentiation and drug screening studies.
HT-29 is a human colorectal adenocarcinoma cell line originally established from the primary tumor of a 44-year-old Caucasian female patient with colon cancer. HT-29 cells exhibit an epithelial-like morphology and grow as adherent monolayers. Cytogenetic analysis shows a hypertriploid karyotype with multiple chromosome aberrations typical of malignant transformation. HT-29 cells harbor key genetic alterations common in colorectal cancers, including mutations in TP53 and APC, deregulated c-myc expression, and BRAF V600E activation, which contribute to proliferative and survival phenotypes. These cells have been used to study epithelial differentiation pathways, intestinal transport mechanisms, and colon tumor biology. HT-29 cell line is also tumorigenic in immunodeficient rodents. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

HT-29 cells are widely used as an in vitro model of human colorectal adenocarcinoma for investigating tumor cell proliferation, differentiation, intestinal transport, and drug response mechanisms. They support studies of chemotherapy sensitivity (e.g., 5-fluorouracil, oxaliplatin), signal transduction pathways, and transporter functions relevant to cancer progression and gastrointestinal biology.
