COLO-320HSR cell line is a human colorectal cancer model widely used for drug testing, tumor biology, and gene regulation research applications.
The COLO-320HSR cell line was derived from the same patient as COLO-320DM but exhibits distinct biological characteristics, growing as adherent epithelial cells rather than floating aggregates. Originating from a metastatic lymph node of a 55-year-old female with poorly differentiated colorectal adenocarcinoma, these cells display classic malignant features including uncontrolled proliferation and anchorage-independent growth. Morphologically, they appear as polygonal epithelial cells forming monolayer sheets. Karyotype analysis reveals chromosomal abnormalities including del(1p), +7, +12, and del(18q) – alterations associated with advanced colorectal cancer. Unlike its parental line, it is adherent and characterized by marker chromosomes with homogeneously staining regions (HSR) and double minutes (DM) at very low levels, reflecting MYC amplification. Gene expression profiling shows TP53 mutation (R273C), and APC mutation. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, syphilis, mycoplasma, fungi, yeast, and bacteria.
