THLE-2 cells are an immortalized human liver epithelial cell line established by infecting primary adult liver cells with recombinant SV40 large T antigen. These cells retain epithelial morphology and grow adherently under standard conditions (37 °C, 5% CO₂). They have surpassed 100 population doublings, display a near-diploid karyotype, and are non-tumorigenic in athymic nude mice. They metabolize pro-carcinogens like aflatoxin B1 and N-nitrosodimethylamine, and express detoxifying enzymes, including epoxide hydrolase, NADPH cytochrome P450 reductase, glutathione S-transferases, and others. These properties make THLE-2 ideal for pharmacotoxicological studies, hepatic metabolism, and hepatocellular carcinoma research. Cells are tested negative for mycoplasma, bacteria, fungi, and cryopreserved for high viability.

THLE-2 cells can serve as useful models for xenobiotic studies. They are commonly used to study insulin pathways and the regulation of hepatic metabolism. These cells are also employed as an lab-based system for drug toxicity tests, allowing researchers to assess the potential adverse effects of drugs and other substances on liver function. Additionally, THLE-2 cells are valuable tools for exploring liver cancer origins and progression, offering insights into the development and progression of this type of liver cancer.
