The TUNEL (TdT-mediated dUTP Nick End Labeling) Apoptosis Detection Kit can be used to detect apoptosis (programmed cell death) in individual cells.
During cell apoptosis, cells activate several DNA endonucleases, which cleave genomic DNA between nucleosomes. When DNA is extracted from apoptotic cells and examined by electrophoresis, a 180-200 bp DNA ladder is observed.
The TUNEL (TdT-mediated dUTP Nick End Labeling) Apoptosis Detection Kit can be used to detect nuclear DNA fragmentation in tissue cells during the late stages of apoptosis. The assay utilizes the incorporation of Fluor 640-12-dUTP into the 3´-hydroxyl (3´-OH) ends of genomic DNA fragments, exposed by the action of terminal deoxynucleotidyl transferase (TdT), allowing detection using fluorescence microscopy or flow cytometry. Fluor 640 is a red fluorescent dye with high stability and strong brightness.
