BT-474 is a human breast ductal carcinoma cell line that was originally isolated from the breast tissue of a 60-year-old Caucasian female patient with invasive ductal carcinoma. BT-474 exhibits epithelial-like, adherent/patchy morphology, growing in compact, patchy, multi‑layered colonies that rarely become fully confluent. The line is highly aneuploid with a modal chromosome number of ~55 and a hypertetraploid karyotype commonly ranging 91–111 chromosomes. These cells robustly expresses estrogen receptor (ER⁺), progesterone receptor (PR⁺), and HER2/neu (ErbB2⁺) and carries a TP53 mutation. BT-474 is tumorigenic in nude mice. These characteristics make it a powerful model for studying hormone-responsive breast cancer.
