Human colon carcinoma. Derived from a colon cancer biopsy of a 55 year old female.
COLO-320 DMF cell line was established from a metastatic lesion of a 55-year-old female patient with poorly differentiated colon adenocarcinoma. Morphologically, COLO-320 DMF cells appear as loosely adherent, round to polygonal cells with a high nuclear-to-cytoplasmic ratio. Karyotypic analysis of COLO-320 DMF reveals complex chromosomal abnormalities and polyploidy, consistent with its malignant phenotype. Gene expression profiling shows co-expression of epithelial markers (CK20) and neuroendocrine markers (chromogranin A, synaptophysin), along with mutated TP53 and elevated MYC oncogene expression. These cells exhibit aggressive malignant characteristics including rapid proliferation, neuroendocrine differentiation, and the ability to form tumors in immunodeficient mice. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, syphilis, mycoplasma, fungi, yeast, and bacteria.

COLO-320 DMF cells are particularly valuable for studying neuroendocrine differentiation in colorectal cancer, a clinically aggressive variant. These cells serve as an excellent model for investigating cancer stem cell properties, drug resistance mechanisms, and metastatic potential. Their unique characteristics make them suitable for research on targeted therapies, immunotherapy response, and tumor heterogeneity. Researchers also utilize COLO-320 DMF cells for studying the epithelial-to-neuroendocrine transition and for developing novel diagnostic biomarkers for poorly differentiated colorectal cancers.
