K-562 is a myelogenous leukemia cell line ideal for studying hematopoietic differentiation, oncogene expression, and blast crisis transformation.
K‑562 is a human chronic myelogenous leukemia (CML) cell line established from the pleural effusion of a 53-year-old female patient in terminal blast crisis and is one of the earliest and most extensively characterized human myelogenous leukemia cell lines. The cells grow in suspension with a rounded, undifferentiated lymphoblast‑like morphology and harbor the Philadelphia chromosome (BCR‑ABL fusion gene), a hallmark of CML oncogenesis. K‑562 cells exhibit multipotential characteristics, retaining capacity for differentiation along erythroid, granulocytic, and monocytic lineages under defined induction conditions, making them a versatile model for hematopoietic differentiation studies. They lack detectable Epstein‑Barr virus sequences and express markers relevant to early hematopoietic progenitors. Evidence for specific metastatic behavior in vivo is not a defining characteristic. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV‑1, HBV, HCV, Syphilis, Mycoplasma, Fungi, Yeast, and Bacteria.

K‑562 is widely used as an in vitro model of human chronic myelogenous leukemia for studies of hematopoietic differentiation, oncogene expression, and signal transduction associated with BCR‑ABL. It supports analyses of erythroid and myeloid lineage specification, investigation of molecular mechanisms underlying blast crisis transformation, and evaluation of therapeutic agents targeting CML progression and leukemia cell survival. K‑562 cells are also used as target cells in natural killer cytotoxicity assays and studies of gene regulation, apoptosis, and chemotherapeutic response.
