MOLM-13
MOLM-13 is a human leukemia cell line derived from the peripheral blood of a 20-year-old Japanese male diagnosed with relapsed acute myeloid leukemia (AML), FAB subtype M5a. The cells display monoblast-like morphology with round to oval shape and grow in suspension culture using RPMI-1640 medium with 10% FBS. When stimulated with IFN-γ alone or combined with TNF-α, the cells can differentiate into macrophage-like cells. MOLM-13 carries a complex karyotype, a FLT3-ITD mutation, and the MLL-AF9 (KMT2A-MLLT3) fusion gene. Its immunophenotype includes CD34+, CD15+, CD33+, CD4+, cyCD68+, while negative for CD13, CD14, CD3, and HLA-DR. The cell line is tumorigenic in immunodeficient mice, although metastatic capacity is not well characterized.

Application

  • MOLM-13 is an important model for studying differentiation and leukemogenesis. For example, used in reverse transcriptase polymerase chain reaction (RT-PCR) analysis, MOLM-13 contains the mRNA of the MLL-AF9 fusion gene, which is a kind of MLL gene rearrangement and related to the leukemia development. MOLM-13 can help identify the genetic profile of human leukemia and identify more genetic markers, such as MLL-AF9 for the diagnosis and prognosis. MLL-rearranged leukemia is hard to treat, MOLM-13 may be used in more scientific research to discover the leukemia mechanisms based on MLL fusions.
