Description

Rat Bladder Smooth Muscle Cells (RBSMC) are primary smooth-muscle cells enzymatically isolated from the detrusor muscle layer of pathogen-free adult Sprague-Dawley rats. They are cryopreserved at an early passage during the log-phase to maintain their contractile phenotype. In vitro the cells display elongated, spindle-shaped morphology, align in parallel bundles and generate spontaneous contractile activity, closely mirroring native detrusor behavior. They express α-smooth-muscle actin (α-SMA). Under pathological conditions, such as mechanical over-distention, oxidative stress or hyperglycemia, the cells recapitulate disease-associated changes, including apoptosis, collagen deposition and altered ion-channel expression resembling overactive or under-active bladder phenotypes. Each lot undergoes rigorous screening and isolation procedures, and is rigorously tested to ensure it is free of contamination from Mycoplasma, Fungi, Yeast, and Bacteria.

  • RBSMCs provide an adaptable platform for dissecting storage–voiding disorders. Researchers employ them to screen pharmacological agents targeting antimuscarinic, β₃-adrenergic or ion-channel modulators, investigate TRP, BK and KV channel signaling that shapes detrusor excitability, study sex-hormone or metabolic influences on contractility and evaluate anti-fibrotic gene or cell therapies aimed at reversing outlet-obstruction or diabetic cystopathy. Their reliable Ca²⁺-imaging, collagen-gel contraction and traction-force assays accelerate high-content drug discovery while reducing reliance on animal strips. In tissue engineering, RBSMCs are used to seed 3D hydrogels or bladder-on-chip devices, supporting development of functional detrusor patches and integrated urothelial-neuronal co-cultures for mechanistic and translational research.
