Leukemia, acute lymphoblastic
CCRF‑SB is a human B‑lymphoblastoid cell line derived from the peripheral blood of an 11‑year‑old Caucasian male patient with acute lymphoblastic leukemia (ALL). The cells exhibit round to polygonal morphology, grow in suspension as lymphoblasts with a doubling time of ~24 hours. They exhibit a normal diploid karyotype (2n=46) and do not synthesize immunoglobulins. CCRF‑SB is of B‑cell lineage, Epstein–Barr virus (EBV)–transformed, and is HLA‑typed (A1, A2, B12, B17, Cw2). This line demonstrates sensitivity to chemotherapeutic agents such as vincristine and methotrexate. It shows strong responses in immune‑related assays and remains non-tumorigenic in nude mice. The cells undergo rigorous screening and isolation procedures, and are rigorously tested to ensure they are free of contamination from HIV-1, HBV, HCV, syphilis, mycoplasma, fungi, yeast, and bacteria.

CCRF‑SB is widely used in immunology, HLA‑profiling, and lymphoid leukemia research due to its B‑cell lineage and EBV transformation. It serves as a crucial tool in drug response studies, particularly with agents like methotrexate and vincristine, including investigations of sensitivity modulation by metabolic cofactors. The cell line supports immune interaction assays, functional genomics, chemoresistance mechanisms, and biomarker discovery. Its extensive omics characterization makes it a powerful tool for transcriptomic and proteomic investigations, while stable suspension growth ensures reproducibility in high-density culture formats.
