KY821 is a human acute myelomonocytic leukemia cell line from peripheral blood with lymphocyte-like morphology and suspension culture properties, ideal for research use.
KY821 is a human acute myelomonocytic leukemia cell line derived from the peripheral blood of a 28-year-old male patient. The cells exhibit lymphocyte-like morphology and grow in suspension culture, representing typical KY821 cell characteristics, and may grow slowly during the initial recovery period after thawing, which should be considered in KY821 cultivation techniques. KY821 cells are characterized by granulocytic maturation and the presence of numerous double minute chromosomes (dmin) derived from chromosome 8, with amplification of the 8q22–24 region containing the MYC oncogene. In addition, KY821 harbors concurrent oncogenic NRAS and TP53 R175H/− mutations, defining key KY821 genetic traits, and sustained expression of mutant p53 is required for maintenance of the leukemic phenotype. These molecular and cytogenetic features contribute to altered stress responses and therapeutic tolerance, making KY821 and its clonal derivatives a relevant model for investigating disease resistance mechanisms in KY821, particularly those associated with dmin-driven MYC amplification and p53 dysfunction.
