THP-1 Monocytic Cells are phagocytic and support monocytic differentiation, inflammation pathway analysis, and immune cell response modeling.
THP-1 is a human monocytic leukemia cell line established from the peripheral blood of a 1-year-old Caucasian male diagnosed with acute monocytic leukemia (AML-M5, FAB classification). The cells grow in suspension with a round morphology and exhibit phenotypic and functional characteristics typical of monocytes. THP-1 cells express surface Fc and C3b receptors but lack both surface and cytoplasmic immunoglobulins. They display positive α-naphthyl butyrate esterase activity that is NaF-sensitive—an enzymatic marker of monocytic lineage—and are capable of phagocytosing latex beads and opsonized sheep red blood cells. Functionally, THP-1 cells secrete lysozyme and can restore Concanavalin A-induced T lymphocyte responses, further supporting their monocyte-like phenotype. This cell line expresses the HLA haplotypes HLA-A2, -A9, -B5, -DRW1, and -DRW2. Cytogenetically, THP-1 exhibits a near-diploid karyotype with multiple structural aberrations, consistent with leukemic transformation. In xenograft models, THP-1 cells demonstrate tumorigenic potential but lack metastatic capacity. THP-1 is widely utilized in immunological research, particularly in studies of the NLRP3 inflammasome, NF-κB signaling, and innate immune responses.
