MBT-2 murine bladder cancer cells grow fast, are metastatic, and express cytokeratin; ideal for syngeneic tumor and bladder cancer drug response studies.
MBT-2 cell line is a chemically induced murine transitional cell carcinoma derived from C3H/He mouse bladder epithelium. This epithelial cell line exhibits a polygonal morphology and forms multilayered colonies in vitro. MBT-2 cells demonstrate a polyploid karyotype, typically ranging from 4N to 16N, and characteristic murine bladder cancer mutations including Hras activation (G12V) and Trp53 alterations. The cells are highly tumorigenic and metastatic in immunocompromised mice, forming invasive, poorly differentiated carcinomas within 2-3 weeks post-implantation. MBT-2 maintains expression of transitional cell markers (cytokeratins, uroplakins) while showing upregulated VEGF and MMP-9 production. As a representative model within bladder cancer cell lines, MBT-2 (Mouse Bladder Tumor-2) is frequently referenced in studies of tumorigenesis and therapeutic screening.

Recommended Culture Medium and Conditions for MBT-2 Cells:
EMEM medium with 10% fetal bovine serum. Some data recommend the MBT-2 to culture in 1640 medium. It depends on your provider laboratory data. Antibiotics such as penicillin-streptomycin are commonly added to prevent contamination.
The doubling time of MBT-2 cells is approximately 1 day, though this can vary depending on culture conditions. Cells are harvested after treatment with 0.25% trypsin and 0.02% EDTA.
In 2000, the cell line was also found to be replicating type C retrovirus contamination.
As with any cell line, there’s a risk of contamination, either with other cell lines or with microorganisms. Contamination can compromise experimental results and lead to erroneous conclusions. Ensuring that the cell line used in experiments is crucial.
